Ok, so everyone is going to need to buy an XBox 360. Then we can all play over the internet too. This video doesn't show it, but I believe IGN has pictures of the new guitar. The old guitar just had 5 buttons at the top, the new one has buttons at the top near the headstock and 5 near the bridge of the guitar (Where you see guitarists strumming). This allows for more complex solos.

Also, if you didn't notice in the video at the end, and in various places in the middle, they allow for improvisation. Although I don't know how well that will work for the guitars, it allows the drummer to put in the custom fills. It's very obvious at the end, but I look forward to this feature the most I think. :)

I can't wait...it looks awesome! The career mode sounds really cool. Start off in a garage and work your way up, purchasing new equipment and such...thats cool! Even creating your own avatar!

EDIT:

Two things I noticed that sound awesome.

1.) You can record your own singles and release them.
2.) Battle of the Bands

These guys manage to add a lot of functionality every release, I really thought the craze was over, but I'm excited all over again.
